In this review, we provide an overview of current in vivo and in vitro models of LDS, discuss how well they recapitulate human disease, and highlight the mechanistic insights they have provided into TGF-β signaling dysregulation, extracellular matrix remodeling, and immune activation, as well as emerging therapeutic avenues.
